Without a doubt, my P220 45 ACP shooting Speer Gold Dot 200gr +P. The most accurate handgun/load combination I've ever shot.
 
My edc "Buddy", a snubby 686 would have no problem with a brain shot at 25yards using SA mode. This sort of scenario is one reason why I prefer a high quality revolver with a superb light crisp SA as well as a good DA. I couldn't do that shot reliably at 25 yards DA.

I would prefer a target hammer, target trigger 6" at 25 yds over a snub, but no matter. BJ GF is good to go.

One nicely placed .530 round ball from my .54 caliber pistol

Failing that I'd use a 230 grain Speer Gold Dot in .45acp from my Colt Series '70

S&W 1911, 230gr Speer Hold Dot. Like others have said, it would be advantageous to have an SA trigger pull for a shot like that.

I've made a similar shot (on steel, 6" circle at 25 yards, timed <5 seconds, from low ready. ironsman.com Try it!) with my M&P 9 as well, but would still prefer the nice single action 1911 trigger pull...
