I had to vote for three; 20, 12, 10.

Similar to @Mopartoolman, we're setup with his and her Mossberg 500s in 12 and 20. Both are setup for defense, securely stored, but readily accessible. (In recent years, better half has transitioned more to PCC though.)

More recently, I've started keeping the 10-gauge SxS in the front of the safe and a five-round box of 00-buck accessible for a quick-ish grab and load. Reason is we've had, in addition to regular concerns about mutants, a surprising amount of wildlife visitors (e.g., black bears, cats, etc.) close to the house in the past year. Figure two 3½" 10-gauge shells, each with 18 pellets of double-ought buck, launched at ~1,300 fps, is going to stop most problems.

I'd like to add a SBS semi-auto in 20 or 12 (probably the latter) to replace mine mentioned above. I'll almost certainly hang on to the pump-action, but it would be good to upgrade soon.

Always gone with 12 as so versatile and ammo is always so easy to find. The Police Mod Win Wife gave me when dating sits with a tube full of 00. Keep a Bandolier of mixed 00 and slugs. Slugs have always been a just in case all hell is breaking loose and had to fire on vehicles. After I added a Shockwave it to is kept full of 00 but side saddle has the rubber less than stuff in case I want to dissuade some hard to convince to leave and not kill.
 
Set my sister up with a 20 gauge cylinder bore. #8 birdshot then #4 buckshot.

Me, 12 ga. It is the universal gauge, can always find slugs, buckshot and birdshot. 20, you have to hunt for or order #4. A shotty is a close range defensive gun. To difficult to feed in multi round scenario, use J frame tactics.
 
I have a semiautomatic 12 gauge FN SLP that I had for years. I use full power 9 pellet 00 buckshot.

I touched off a couple of those dragon fart .410 rounds once without realizing what they were.
Anemic sparklers . I've seen more heat generated by those novelty birthday candles.
 
My defense shotgun, which is the gun beside my bed, is a 7 shot 12 gauge pump with a 20" barrel.
However, my sister, who is not large or highly experienced, has a 20 gauge Wingmaster that I had the barrel shortened to 18". (Well, she will have it once her new house is built...) I bought some Federal defense #4 buck for that, too.
Shotgun and buckshot. Fight stopper in any gauge.
 
I cast votes for four: .410 bore and 28, 20 and 12 ga. Because I hand load solid projectile loads for all four. And of course I have various shot loads for them all. Might as well be versatile.
 
12ga. in a benelli supernova tactical. Double ought in the tube with a sleeve of 5 on the buttstock. Three 00 in front, two slugs behind.
If that doesn't do it, I'm in way over my head!

Doesn't it make more sense for you both to have the same for commonality/compatibility whether it be 12 or 20?
 
I picked 12 ga, because there are sooooo many specialty round available. Buckshot, mini shells of buck, slugs, etc... try and find those in any but 12 ga.
I can find light loads, featherlight Win AA, mini shells all for those that are recoil shy. these are not available in anything but 12 ga.
16,20,28, and 410. all have their places. there is nothing easier to carry all day than a 28 ga. But how many will be carrying a HD gun all day? DR
